Kisspeptin is a family of neuropeptides encoded by the KISS1 gene, acting as the most potent known endogenous activator of the hypothalamic-pituitary-gonadal (HPG) axis. What Is Kisspeptin?
The KISS1 gene encodes a 145-amino acid precursor protein (prepro-kisspeptin) that is cleaved to produce several bioactive fragments. The primary research forms are:

Mechanism of Action
KISS1R (GPR54) Activation
Kisspeptin binds to KISS1R, a Gq/11-coupled receptor expressed on GnRH neurons in the hypothalamus. KISS1R activation triggers:
- Phospholipase C (PLC) activation
- IP3-mediated calcium release
- Protein kinase C (PKC) activation
- Depolarization of GnRH neurons
- Pulsatile GnRH release into the hypothalamic-pituitary portal circulation
GnRH Pulse Generation
Kisspeptin neurons in the arcuate nucleus (ARC) are the primary drivers of pulsatile GnRH secretion — the fundamental signal that drives LH and FSH pulsatility from the pituitary. The ARC kisspeptin neurons co-express neurokinin B (NKB) and dynorphin, forming the "KNDy" neuron population that generates the GnRH pulse generator.
Kisspeptin neurons in the anteroventral periventricular nucleus (AVPV) mediate the preovulatory LH surge in females, driven by positive estrogen feedback.
Downstream HPG Axis Activation
Kisspeptin → GnRH → LH + FSH → Gonadal steroidogenesis (testosterone, estradiol, progesterone)

Negative and Positive Feedback Integration
